What is MBRT?

MBRT means Methylene Blue Reduction Test. It is used in dairy quality control to estimate the microbial quality of milk. The test works on a simple scientific idea: bacteria consume dissolved oxygen during growth. As oxygen decreases, methylene blue changes from blue to colorless.

Key message: The faster the blue color disappears, the higher the microbial activity and the poorer the milk quality.

Scientific Principle

Methylene blue acts as a redox indicator. In good quality milk, oxygen remains available for a longer time, so the dye stays blue. In poor quality milk, bacteria rapidly use oxygen and produce reducing substances, causing faster decolorization.

Milk bacteria grow → Oxygen decreases → Reducing environment forms → Blue dye becomes colorless → Quality is interpreted

Standard Procedure

1. Sample

Take 10 mL well mixed milk in a clean sterile test tube.

2. Dye

Add 1 mL methylene blue solution and mix gently.

3. Incubate

Keep at 37°C and avoid unnecessary shaking.

4. Record

Note the time required for complete loss of blue color.

Interpretation of Results

Grade Reduction Time Milk Status Action
Excellent More than 8 hours Low microbial activity Accept
Good 5 to 8 hours Acceptable quality Accept with monitoring
Fair 2 to 5 hours Hygiene concern Corrective action
Poor Less than 2 hours High spoilage risk Reject or investigate

Industrial Quality Control Workflow

Milk Collection → Chilling → Transport → Reception → MBRT Screening → Decision → Processing → Storage → Distribution

Advantages

Simple, low cost, quick screening test for raw milk reception and supplier quality monitoring.

Limitations

Does not identify specific pathogens, adulterants, toxins, antibiotics or legal microbiological compliance.

Best Use

Use as a first screening tool with standard plate count, coliform count and temperature monitoring.

MBRT Cannot Replace These Tests

For legal, export, public health or regulatory action, MBRT should be supported by confirmatory laboratory testing.

Standard Plate Count
Coliform Count
Pathogen Testing
Antibiotic Residue Test
Adulteration Analysis
Aflatoxin M1 Testing

Expert Conclusion

MBRT is not a final safety test. It is a practical screening tool that helps dairy professionals judge raw milk hygiene, handling quality and microbial activity before processing. The strongest dairy control system combines MBRT with chilling control, supplier monitoring, sanitation verification and confirmatory microbiological testing.
